The multiresidue determination of pesticides in barley and malt by the LC/MS method with QuEChERS extraction

The topic of this thesis is the determination of pesticides in barley and malt. Barley is one of the second most cultivated cereals in the Czech Republic. In order to achieve higher yields, higher grain quality, and avoid losses, plant protection products - pesticides - are used. The disadvantage of their use is their negative impact on human and animal health and the environment. Residues of pesticides and their metabolites can remain in a food, so it is a good idea to monitor their content. There are various options for determination, but recently the QuEChERS extraction method has been used, which speeds up the whole sample preparation process. The practical part summarizes the optimization and validation of the QuEChERS method and the subsequent quantification of analytes by UPLC-MS/MS. The content of 148 pesticides in 30 barley and 20 malt samples from the 2021 harvest in the Czech Republic was analysed.

The aim of the bachelor thesis was to quantify the effect of the form of nitrogen fertilization on the yield and quality of barley. For this purpose, a pilot experiment was established on a family farm near Tábor. The experiment was based into two variants: variant I using a solid form of nitrogen fertilizer - ammonium nitrate and variant II using a liquid form of nitrogen fertilizer - urea. The nitrogen dose for both variants was 100 kg.ha-1 No. b. The monitored indicators were the number of ears per m2, the number of grains in the ear, 1,000 K weight, the proportion of front grain, the content of crude protein and germination. In the number of ears per m2, the number of grains, the proportion of front grain, the crude proteinand germination, the variant using ammonium nitrate reached higher values. The 1,000 K weight value was the same for both variants.

This bachelor thesis compares grain production efficiency in conventional and organic agricultural settings. The testing objects are wheat, barley, and rye. In the practical section of this work, the gained data are analyzed from the perspective of revenue, cost, and price from the years 2010 to 2015. From the research outcome the organic agriculture can ensure only about one half of the conventional agricultural production; on the other side, the price reaches 1.5 times higher values in the organic produced crops. The cost structure is different. Material cost appears higher under the conventional conditions. Manual labor and machinery use increase under the organic circumstances. Conventional agriculture shows more beneficial in reflection of the contribution margin and return on cost (ROC) calculations. However, when production subsidies are incorporated, organic agricultural production reaches more favorable economical results.
